Are you using a Prolific USB-to-Serial adapter?  Check the driver name in
the Windows Device Manager.  Many Prolific adapters do not work.  FTDI
adapters such as the GearMo brand are much more reliable than Prolific.

The symptom you describe is consistent with my observation that some
USB-to-Serial adapters require RTS or DTR to be set to "Always ON" or
"High" to supply consistent power to the adapter before they will properly
poll the radio.
